Video Summary
The speaker described a study of allergen-specific tissue-resident memory CD4 T cells in a mouse model of house dust mite–induced airway allergy. Using lineage tracing and single-cell RNA sequencing, the team found heterogeneous lung TRM2 populations, including a novel Th2+17 subset that co-expresses GATA3 and RORγT/IL-17. These cells persisted in the lung for up to one year, were antigen-specific, and could arise from Th2 cells after rechallenge. Similar Th2+17 cells were also detected in asthma patient lung data, suggesting they may contribute to severe, steroid-resistant asthma and represent a potential therapeutic target.
